In vitro Culture of Bulbil Scale of Lilium lancifolium




Abstract
The study was conducted in order to obtain basic informations for mass production of Lilium species relatively difficult to propagate and clarified effect of auxins for proliferation of bulblet and growth of seedlings, when bulbil tissues were cultured in Murashige and Skoog¢¥s medium supplemented with various auxin concentrations.
The results obtained were summarized as follows.
Bulblet formation was highly accelerated from basal part of bulbil tissue between second week and third week after incubation and bulblet from distal part was formed continuously from second to seventh week, but average ratio of bulblet formation was higher from basal part than that from distal part.
Ratio of bulblet formation from basal part was the best in the control plot and in the media added with 0.01ppm NAA, 0.01, 0.1, and 1.0ppm IAA, 0.01 and 0.1ppm IBA, but that from distal part, in the medium added with 0.1ppm IAA only.
No. of bulblets originated from basal part were the more numerous than those from distal part in all the media tested, especially those were the most in 1.0ppm IBA added medium.
Bulb width and fresh weight of bulblet showed the highest increased tendency in 0.1ppm NAA added medium. Bulblet height was the highest in 1.0ppm IBA added medium. Leaf length was the longest in 1.0ppm NAA added medium. and root length, in 0.01ppm IBA added one.
